But not all tests are the same. Different methods measure different things: airborne spores, surface growth, settled dust, or chemical byproducts.

Here’s a clear, practical guide to the most common mold testing methods, plus an explanation of specialized tests like ERMI, HERTSMI, actinomycetes, endotoxin, and beta glucan. Understanding what each test shows helps you get the full picture of your home’s safety.

1. Air Sampling

What it does: A pump draws air through a special cassette that traps airborne mold spores and particles. The sample is then analyzed under a microscope or by culture.

Best for: Detecting current airborne mold levels and identifying the types of mold present in the breathing air.

Strengths: Quick, non-invasive, and shows what you’re actually inhaling right now.

Limitations: Results can vary with air currents, HVAC use, or recent cleaning. It may miss hidden mold that isn’t actively releasing spores.

When to use: As part of a whole-home assessment, especially in rooms where people spend the most time.

2. Tape Pulls / Surface Sampling

What it does: Clear tape or a swab is pressed onto a surface (wall, ceiling, floor) to lift visible or suspected mold growth for lab analysis.

Best for: Confirming visible mold growth and identifying the exact species on surfaces.

Strengths: Simple and inexpensive; gives direct evidence of active growth.

Limitations: Only tells you what’s on that specific spot — it doesn’t show hidden mold inside walls or under floors.

When to use: When you see discoloration, staining, or suspected growth.

3. Dust Sampling

What it does: A vacuum with a special collection device samples settled dust from carpets, floors, or furniture. The dust is analyzed for mold DNA or particles.

Best for: Revealing long-term, accumulated contamination that has settled over time.

Strengths: Captures what’s been building up in the home, even if mold is no longer actively growing.

Limitations: Doesn’t show current airborne levels.

When to use: To assess overall home contamination and secondary contamination of belongings.

4. MVOCs / VOCs Testing

What it does: Air is sampled and analyzed for volatile organic compounds (VOCs) and microbial VOCs (mVOCs) — the chemical gases produced by mold and bacteria.

Best for: Detecting hidden mold that isn’t producing many visible spores (e.g., inside walls or HVAC systems). The characteristic “musty” smell is often from mVOCs.

Strengths: Can find mold growth that air sampling or tape pulls might miss.

Limitations: More expensive and results can be influenced by other household chemicals or outdoor air.

When to use: When you have a persistent musty odor but no visible mold.

Specialized Tests: ERMI, HERTSMI, Actinomycetes, Endotoxin & Beta Glucan

These are advanced lab tests performed on dust samples and are especially valuable in water-damaged homes.

  • ERMI (Environmental Relative Moldiness Index): A DNA-based test that analyzes dust for 36 specific mold species. It compares your home’s mold levels to a national database and gives a single score. Higher scores indicate greater mold burden and health risk.
  • HERTSMI-2: A simplified version of ERMI that focuses on the 5 most problematic molds. It’s commonly used to determine whether a home is safe to return to after remediation (a score below 10 is generally considered acceptable).
  • Actinomycetes: This test looks for specific bacteria that thrive in the same wet conditions as mold. Elevated levels are a strong indicator of chronic moisture problems and can contribute to inflammation.
  • Endotoxin: Measures bacterial toxins (from gram-negative bacteria) commonly found in water-damaged buildings. Endotoxins are highly inflammatory and can worsen symptoms even when mold levels are moderate.
  • Beta Glucan: Detects fungal cell wall components. Elevated levels indicate mold presence and can trigger immune reactions and inflammation in sensitive people.

Why Do We Want All of This Information?

No single test tells the whole story. A home can have:

  • High airborne spores but low settled dust
  • Hidden mold that only shows up on MVOC testing
  • Bacterial co-contamination (actinomycetes + endotoxin) that air sampling misses

Combining air sampling, dust sampling, surface sampling, and specialized tests like ERMI/HERTSMI gives you a complete picture of:

  • Current exposure risk
  • Hidden contamination
  • How contaminated your belongings and HVAC system may be
  • Whether the home is safe for sensitive occupants
